The first meeting of the Executive Board of the Monroe County Birth Control League was held on November 28, 1932. They opened their first birth control clinic two years later in the basement of the First Unitarian Church's Gannett House in downtown Rochester.

During the 1940s the name of the organization was changed to the Monroe County League for Planned Parenthood. In 1955 the name was changed to Planned Parenthood League of Rochester and Monroe County then to Planned Parenthood of Rochester and Monroe County. In 1986 the organization moved to their fifth location, a new facility at 114 University Avenue. At that time the name was changed to Planned Parenthood of Rochester & the Genesee Valley. The organization is now called Planned Parenthood of the Rochester/Syracuse Region.
